...reflected off the object. This theoretical (and practical) limit is known as the diffraction limit.

In order to transcend this limit to image much smaller features and objects, such as proteins or DNA, researchers have developed various techniques, including the use of longer-wavelength electromagnetic energy (UV and X-rays), contrast stains, fluorescent dyes, laser light sources, or electron beam sources. For example, atomic force microscopy (AFM) uses a cantilevered probe to scan the surface of a nanometer-sized object, while a laser detector records the probe’s movements to calculating the atomic forces within that object.

Nearfield scanning optical microscopes (NSOM) produce images of surface plasmons. Multiphoton microscopes allow deep tissue imaging of cellular structures such as neurons, without tissue slicing. Stochastic optical reconstruction microscopy (STORM) technology enhances resolution to near-molecular levels. Microscopy can also be combined with spectroscopy; for example, Raman microscopectroscopy uses Raman scattering for non-destructive chemical analysis of microscopic objects, such as proteins and cells.

Accessories for microscopy include objective lenses, microscope software and digital control units, microscope mounted CCD cameras to record real-time images of living cells, transilluminators, and microimaging systems for surgical imaging.
